The generation of transgene Compound chromosome fly lines

Description
This thesis describes the establishment of a viral tranfection system - phiC31 - into the compound chromosomebackground. Utilising cold-shock induced non-disjunction in females, it was possible to viably breed flies bearing a specific attP-landing site and compound C(2)v males. When those flies get injected with a construct containing the counterpart attB, integration will soley occur at the previously introduced landing site, at up to five-fold higher rates. The merger of phiC31-system and compound chromosomes simplifies transgenesis and enables highly comparable and reproducible investigation of early embryogenesis events.
